In order to better understand template enrichment we would like to explain the difference between target and template enrichment.
An NGS target normally means the DNA region of interest that will be sequenced. This can be a gene, a part of a gene, or a larger genome fragment. Often, target amplification is performed by PCR using specific NGS gene panels.
An NGS template consists of already processed DNA fragments that later go on the sequencing platform. A template can also denoted as part of the NGS library. This can be for example processed DNA fragments from a sequencing library coupled to magnetic beads (Ion Torrent) or DNA fragments bound on a flow cell (Illumina). Depending on the sequencing platform, these templates normally undergo an additional amplification step prior the sequencing process.
